Zeiss LSM 710 Confocal and Elyra S.1 Structured Illumination Super Resolution Microscope

The Zeiss LSM 710/Elyra S.1 is a single microscope stand that functions as both a laser scanning confocal and a structured illumination super resolution microscope.

The shared Zeiss Axiovert microscope stand includes:

  • Objectives:
    • EC Plan-Neofluor 10X; NA = 0.30, WD = 5.2mm
    • Plan-Apochromat 20X; NA = 0.80, WD = 0.55mm
    • Plan-Apo 40X (oil); NA = 1.4
    • Plan-Apo 63X (oil); NA = 1.4
    • Water 63X (oil); NA = 1.2
  • Incubation: The microscope has incubation controls for both temperature and CO2 for live cell imaging.

The Zeiss LSM 710 Confocal Microscope can be used for fluorescent imaging of a variety of biological and material samples.  The instrument provides high resolution images using a variable pinhole for optical 'Z' sectioning and laser scanning.  The system is equipped with the following excitation wavelengths: 405nm, 488nm, 514nm, 561nm and 633nm.  The filterless scanhead allows for tunable emission detection of a wide array of fluorescent molecules.

Advanced imaging techniques including spectral imaging, FRET and FRAP are availabe on this instrument.

The Zeiss Elyra S.1 Structured Illumination Super Resolution system achieves a lateral resolution of ~100nm and an axial resolution of ~200nm through the use of SR-SIM.  This system has the ability to penetrate up to 20um into the tissue and can acquire 3 Z-stack, multi-channel SR-SIM data with up to 4 channels.  The system is equipped with 4 excitation laser wavelengths for standard fluorophores (405 nm, 488nm, 561 nm and 633nm) and comes with emission filters for DAPI, GFP, RFP and Alexa 633.

Compound Microscopes

Zeiss Axiovert 200M Fluorescent Microscope

The Zeiss Axiovert 200M is a widefield fluorescent microscope fitted with a 100W Hg lamp variable intensity power supply.  The instrument has fluorescence filter cubes for a wide range of dyes including DAPI, CFP, FITC, GFP, YFP, TRITC, CY5 and CFP/YFP FRET.  Brightfield and DIC imaging for high magnification is also availalbe.

Nikon 90i Stereology System

The Nikon 90i Stereology microscope is run with MBF Stereology software and is fitted with an Excite fluorescent lamp and Halogen lamp for brightfield imaging.  The instrument has fluorescence cubes GFP and RFP.  Images are captured on a color camera and are analyzed using MBF Stereology analysis tools.

Leica DM2500 Compound Light Microscope and Leica S6D Stereoscope

The Leica DM2500 Compound Light Microscope is used for general transmitted light images.  The microscope shares a Leica DFC color camera with the Leica S6D stereoscope.  Several objectives are fitted with optics for DIC and phase.

The Leica S6D Stereo Microscope is commonly used for dissection, inspection and documentation of cryostat and ultramicrotome sections, but is also useful for large samples or samples with non-typical mounting.  The microscope has a zoom range of 0.63X to 4X and is fitted with white light for reflection or transmission imaging.  Images can be acquired using a Leica DFC color camera.

Fluorescent Stereoscope

Leica MZ16

The Leica MZ16 Fluorescence Stereomicroscope is designed for acquisition of fluorescent images of many dyes including DAPI, FITC, TRITC, CFP and YFP at long working distance and low magnification (4X - 20X).  The instrument has a black and white camera for fluorescent image acquisition and a color camera for imaging of vital dyes.  The stage is motorized and is capable of X, Y and Z montage imaging.
